چکیده

Identification of a Mutation in COL4A5 Causative for X-linked Alport Syndrome in the Domestic Dog and Analysis of Gene Expression in the Kidneys of Affected and Nonaffected Siblings. (December 2003) Melissa Luanne Cox, B.Sc., Dalhousie University Chair of Advisory Committee: Dr. Keith E. Murphy The domestic dog, Canis lupus familiaris, plays many roles in the lives of humans. Additionally, the dog is recognized for its potential as a model for many human hereditary diseases. Thus, the genetics and genomics of the dog are being studied extensively in order to facilitate its use as a model, as well as to help the dog for its own sake. As part of this research effort, our laboratory has added type I markers (i.e., the acidic and basic keratins, c-kit, type I and IV collagens, and the gene encoding uromodulin) to the emerging map of the canine genome. The mapping of genes, particularly those in large gene families such as the collagens, is valuable because it rapidly increases the density of gene loci on the map and provides insight regarding conservation of synteny between the dog and other mammals. The major focus of work reported here is the genetics of X-linked Alport syndrome (XLAS), a terminal renal disease that affects the human and the dog. The disease results from mutations in COL4A5, a type IV collagen gene. Reported here are the 1) sequencing and mapping of the canine cDNA encoding uromodulin, 2) mapping of the type I and type IV collagen

A Large Deletion in the NSDHL Gene in Labrador Retrievers with a Congenital Cornification Disorder

In heterozygous females affected by an X-linked skin disorder, lesions often appear in a characteristic pattern, the so-called Blaschko's lines. We investigated a female Labrador Retriever and her crossbred daughter, which both showed similar clinical lesions that followed Blaschko's lines. The two male littermates of the affected daughter had died at birth, suggesting a monogenic X-chromosomal...

Gains from diversification on convex combinations: A majorization and stochastic dominance approach

By incorporating both majorization theory and stochastic dominance theory, this paper presents a general theory and a unifying framework for determining the diversification preferences of risk-averse investors and conditions under which they would unanimously judge a particular asset to be superior. In particular, we develop a theory for comparing the preferences of different convex combination...

Improved immunogenicity of tetanus toxoid by Brucella abortus S19 LPS adjuvant.

BACKGROUND Adjuvants are used to increase the immunogenicity of new generation vaccines, especially those based on recombinant proteins. Despite immunostimulatory properties, the use of bacterial lipopolysaccharide (LPS) as an adjuvant has been hampered due to its toxicity and pyrogenicity. Brucella abortus LPS is less toxic and has no pyrogenic properties compared to LPS from other gram negati...

Steady electrodiffusion in hydrogel-colloid composites: macroscale properties from microscale electrokinetics.

A rigorous microscale electrokinetic model for hydrogel-colloid composites is adopted to compute macroscale profiles of electrolyte concentration, electrostatic potential, and hydrostatic pressure across membranes that separate electrolytes with different concentrations. The membranes are uncharged polymeric hydrogels in which charged spherical colloidal particles are immobilized and randomly d...

Perturbative Analysis of Dynamical Localisation

In this paper we extend previous results on convergent perturbative solutions of the Schrödinger equation of a class of periodically timedependent two-level systems. The situation treated here is particularly suited for the investigation of two-level systems exhibiting the phenomenon of (approximate) dynamical localisation. We also present a convergent perturbative expansion for the secular fre...
